Kinetic Modelling of Oligonucleotide Detritylation

High Throughput Experimentation

Program Overview

  • Use of kinetic modeling with oligonucleotides.
  • Nuances of liquid phase oligonucleotide kinetics.
  • Combining high throughput experimentation with simple kinetic modeling for strategic experimentation.

This presentation will discuss two examples of using kinetic modeling to advance the development of the detritylation stage of a liquid phase oligonucleotide synthesis process. Case study 1 will explore developing a kinetic model that can cope to various changes in process parameters including nucleotide type, acid concentration and oligonucleotide concentration. Case study 2 will explore the use of kinetics and high throughput experimentation to shortlist a selection of reagents and solvents for detritylation.
